Thiamine occurs in cells principally in its active coenzyme form called thiamine pyrophosphate (TPP, cocarboxylase). Thiamine, in the form of thiamine pyrophosphate, plays an essential role as a cofactor in key reactions in carbohydrate metabolism. It is also involved in the metabolism of branched-chain amino acids and may have non-coenzyme (non-cofactor) roles in excitable cells. Thiamine has been shown to possess antioxidant, erythropoietic, cognition-and mood-modulatory, anti-atherosclerotic and detoxification activities.
Riboflavin has antioxidant activity. It may have activity in the prophylaxis of migraine headaches and may have activity against oesophageal cancer. It has anti-atherosclerotic activity and putative antimalarial activity. Riboflavin activates vitamin B6 and folate, is a coenzyme in the respiration enzyme system and is required for the maintenance of the mucosal, epithelial and eye tissues, myelin sheath and the redox and respiratory enzymes.
Niacin, via its metabolites, is involved in a wide range of biological processes, including the production of energy, the synthesis of fatty acids, cholesterol and steroids, signal transduction, the regulation of gene expression and the maintenance of genomic integrity. Nicotinic acid, in pharmacological doses, is used as an anti-hyperlipidemic agent. Niacin stimulates tyrosine hydroxylase, gastric secretion and bile secretion.
Vitamin B5 is involved with the synthesis of acetylcholine, antibody production, cholesterol, cortisone and lipid metabolism. Pantothenic acid is a precursor to coenzyme A. Deficiency symptoms as related to vitamin B5 metabolic action occur as cardiovascular disorders – disturbances of lipid and carbohydrate metabolism, digestive disorders – decreased bile acid production and atrophy of intestinal mucosal, and neuromotor disturbances – decreased synthesis of acetylcholine.
Pyridoxine is involved with the transamination reactions which produce substrates to feed the Krebs cycle and hence oxidative metabolism. Pyridoxine has been used therapeutically to treat ‘hyperventilation syndrome’, a related stress-induced disorder characterised by increased respiration, anxiety, muscular hypertonia and is required for the metabolism of neurotransmitters and homocysteine. Vitamin B6 regulates estrogen metabolism, elevated serum estrogen has been implicated in depression and anxiety states.
Vitamin B12 and Folate help reduce production of the toxic amino acid homocysteine, which has been implicated in the aetiology of a number of chronic disorders, including cardiovascular disease, depression and dementia. Folate is required for the production of SAMe and the metabolic conversion of dopamines.
Biotin is essential for the maintenance of skin, hair, sebaceous glands, bone marrow and sexual glands, metabolism of fat, protein and sugars, cellular growth, liver function and reduces cholesterol plaques in blood vessels. Biotin may also reduce the conversion rate of candida albicans.

Oral contraceptives and the drug Doxorubicin may decrease serum riboflavin activity.
Niacin may cause skin flushing in some individuals. This may be unpleasant, but is beneficial from a health perspective. If flushing is inappropriate, B complex should be consumed after food and diluted in juice or water.
